In this way, we will find those genes acutely regulated by insulin and those whose regulation is perturbed in an insulin-resistant state. |

| My aim is to understand the intracellular pathways by which insulin carries out its metabolic effects and the mechanism by which these pathways are down-regulated in insulin-resistant states. To address these questions, we are taking advantage of GNF's leading technology in the areas of genomics nutritional supplement and proteomics.. Phosphoproteomics is the study of the phosphorylation state of the entire complement of cellular proteins. Coupling phosphotyrosine immunoprecipitation and immobilized metal affinity chromatography with mass spectrometry allows us to take an unbiased look at tyrosine phosphorylation nutritional supplement events in the cell. We are using this nutritional supplement phosphoproteomics approach, to look at signaling events downstream of insulin binding to its receptor, both in vitro and in vivo. Preliminary results from Chinese hamster ovary (CHO) cells over expressing the insulin receptor show us some of the proteins we expect to be tyrosine phosphorylated in response to insulin, like insulin receptor and IRS1. |
